Abstract

Summary of the research findings

Here, we describe a protocol to analyze ancient DNA (aDNA) from skeletal remains discovered at Quarto Cappello del Prete, a rural Imperial Roman site located in the Roman suburbium (1st-3rd centuries CE). The protocol comprises guidelines to extract aDNA from archaeological human remains, construct NGS libraries, and perform computational analyses for population genetics studies.
